Quality of Etawah Mutant Goat Cement in Modification of Tris Diluents with Trehalosa and Rafinosa

This study aimed to compare the effectiveness of trehalose and rafinose supplementation in Tris yolk and Tris soya diluents in optimizing the quality of the liquid semen of Etawah Breeders (PE) goats. Semen was collected from three adult male PE goats using an artificial vagina. Semen is evaluated and divided into six parts. Then diluted using Tris yolk (TKT) and Tris soya (TS) diluents by supplementing 50 mM trehalose and raffinose, liquid cement is stored in the refrigerator (5 ° C). Evaluation of spermatozoa motility, live spermatozoa and intact plasma membrane (MPU) were carried out every 12 hours until motility reached 50%. The results showed that the progressive motility of spermatozoa in TKT diluents with trehalose and rafinose supplementation could last up to 50% for 72-84 hours, better than TS diluents which persisted for 48-60 hours (P <0.05). The best diluents were shown by TKT with trehalose supplementation (52.82 ± 3.21%) lasting up to 84 hours compared to raffinose supplementation (52.78 ± 4.41%) and controls (51.78 ± 4.86%) to 72 hour. Meanwhile in TS diluents supplemented with rafinose (52.78 ± 4.41%) lasted up to 60 hours compared with trehalose (53.33 ± 3.54%) and controls (51.11 ± 4.86%) to 48 hour. All spermatozoa thinners lived 6-9% higher than the percentage of spermatozoa motility, while the MPU percentage value was almost the same as the percentage of spermatozoa motility. It can be concluded that for the preservation of liquid semen the best PE thinner goats are TKT with trehalose supplementation, and in TS thinners better supplemented with raffinose.
