Non-Invasive Mitochondrial DNA (MtDNA) BILOU Analysis Technique (Hylobates klossii, Miller 1903) Through Polymerase Chain Reaction

Abstract

Abstract: Non-invasive analysis techniques for mitochondria bilou DNA (Hylobates klossii) with Polymerase Chain Reaction (PCR) are molecular techniques that can support animal welfare. Non-invasive analysis of stool samples can determine DNA samples from the control area in 12S rRNA. Fecal samples were collected from three different islands (Sipora, Siberut and Pagai) with a total of 39 samples extracted for DNA. Analysis was carried out at the LIPI Zoological Genetics laboratory in Cibinong in July to August 2005. The PCR test results used by Qiagen QIAamp ® DNA stool mini kit and electrophoresis can show that DNA samples can be displayed even in the lowest concentration of spectrophotometer measurements. Thus, this can be a new reference in future research as an indicator of conservation management in accordance with good animal welfare regulations. Polaroid photos from rRNA 12S (L = 1091, H = 1478) taken with UV transluminators and Polaroid MP4 cameras from PCR products with annealing were used at 50 ° C in 30 seconds, showing results from DNA electrophoresis marker bands ranging from 750 to 815 pb DNA ladder.
